Luteolin inhibits NLRP3 inflammasome activation via blocking ASC oligomerization

چکیده

The NLRP3 inflammasome is a caspase-1 containing multi-protein complex that controls the release of IL-1β and plays important roles in innate immune response. Since implicated pathogenesis variety diseases, it has become an increasingly interested target developing therapies for multiple diseases. We reported current study to determine how luteolin, natural phenolic compound found many vegetables medicinal herbs, would modulate both vivo vitro settings. First, we high-fat diet upregulated mRNA expression components Asc Casp1 adipose tissue ovariectomized mice, which were greatly reduced by dietary supplementation with luteolin. Of note, correlated levels Adgre1 encoding F4/80, established marker mature macrophages. also demonstrated luteolin inhibited inflammasome-derived activation secretion J774A.1 macrophages upon diverse stimuli including ATP, nigericin, or silica crystals. Luteolin step interfering ASC oligomerization. Taken together, these findings suggest may suppress induction process thus potentially be protective against NLRP3-mediated inflammatory

Genipin inhibits NLRP3 and NLRC4 inflammasome activation via autophagy suppression

Inflammasomes are cytoplasmic, multiprotein complexes that trigger caspase-1 activation and IL-1β maturation in response to diverse stimuli. Although inflammasomes play important roles in host defense against microbial infection, overactive inflammasomes are deleterious and lead to various autoinflammatory diseases. In the current study, we demonstrated that genipin inhibits the induction of IL...

Quercetin Inhibits Inflammasome Activation by Interfering with ASC Oligomerization and Prevents Interleukin-1 Mediated Mouse Vasculitis

Interleukin-1β (IL-1β) is a highly inflammatory cytokine that significantly contributes to both acute and chronic inflammatory diseases. The secretion of IL-1β requires a unique protease, caspase-1, which is activated by various protein platforms called inflammasomes. Data suggests a key role for mitochondrial reactive oxygen species for inflammasome activation. Flavonoids constitute a group of...

Fluoxetine Inhibits NLRP3 Inflammasome Activation: Implication in Depression

BACKGROUND Emerging evidence indicates that NLRP3 inflammasome-induced inflammation plays a crucial role in the pathogenesis of depression. Thus, inhibition of NLRP3 inflammasome activation may offer a therapeutic benefit in the treatment of depression. Fluoxetine, a widely used antidepressant, has been shown to have potential antiinflammatory activity, but the underlying mechanisms remain obsc...

Celastrol ameliorates inflammation through inhibition of NLRP3 inflammasome activation

Celastrol exhibits potential anti-inflammatory activity in a variety of inflammatory diseases, but the mechanism remains poorly understood. Activation of NLRP3 inflammasome is involved in multiple inflammatory diseases. Here, we show that celastrol abolishes the NLRP3 inflammasome activation, inhibits subsequent caspase-1 activation and IL-1β secretion both in vitro and in vivo. Notably, interr...

Dietary PUFAs attenuate NLRP3 inflammasome activation via enhancing macrophage autophagy

Dietary PUFAs reduce atherosclerosis and macrophage inflammation, but how nucleotide-binding oligomerization domain leucine-rich repeat-containing receptor protein (NLRP3) inflammasome activation and autophagy influence PUFA-mediated atheroprotection is poorly understood. We fed Ldlr-/- mice diets containing 10% (calories) palm oil (PO) and 0.2% cholesterol, supplemented with an additional 10% ...
